The mass spectrometry (MS) resources on campus aim to provide researchers with state-of-the-art techniques and equipment to characterize biomolecules including proteins and peptides (proteomics, sugars including glycoproteins (glycomics), metabolites (metabolomics) and lipids (lipidomics). A number of state of the art instruments for the separation of biomolecules and their analysis by mass spectrometry based approaches are available. In addition to providing analytical services, the mass spectrometry facility on campus provides training to on-campus scientific staff on the use of this technology as well as develop new analytical methods required to facilitate on campus research.
 
Lipidomics:
Facility is driven to establish a hyphenated Mass Spectrometry based Lipidomics platform in basic biology, clinical studies and biotechnology at Bangalore Life Sciences Cluster (BLiSc) affiliated institutes; NCBS, CCAMP and InStem. The platform undertakes clinical, biological, chemical, ecology studies of lipidomics/ metabolites for BLiSc community and external researchers. Direct infusion as well as LC-MS/MS methods using High Resolution Mass Spectrometry (HRMS) like Orbitrap technology (Q-Exactive, Orbitrap XL) are available. The facility also has highly sensitive triple quad mass spectrometers such as Agilent 6495 for targeted lipidomic.
Identification and quantification of known and unknown Lipids/metabolites in biological fluids such as plasma, urine, saliva and tissue using Data dependant (DDA) and data independent (DIA) analysis for the estimation of lipids and other metabolites. Comprehensive lipid profiling and quantitation is a system-based study of all possible lipid classes in the biological system. Phosphoinositides measurements in cell signaling.
Lab is also well equipped with other sample preparation instruments such as solid phase extraction (SPE) and liquid-liquid extraction (LLE).
Well versed training program is available for the internal as well as external researchers throughout the year.

Instruments for Proteomics / Protein Characterization

     
Thermo Q-Exactive - The combination of LC separation, high resolution MS and MS2 analysis should provide unambiguous and precise characterization of different classes of lipids in biological samples. Q-exactive MS with its mass resolution of 140,000 at 400 m/z and mass accuracy of 1-2 ppm is optimized for lipidomics and metabolomics experiments.  HCD fragmentation using stepped normalised collision energies is an excellent tool for identification and quantitation of different classes of lipids.       Sciex QTRAP 6500 MS/MS - One of the best is class triple-quadrupole MS instrument for the targeted quantitative analysis with MS3 fragemntation that enable for identification as well. It also equipped with the differnetial ion mobility technology.
         
     
Agilent nano HPLC - This LC system with flow rate ranges from 100 nl/min to 1ul/min is suitable for high sensitivity MS detection and 2 dimensional separation.       Advion Triversa NanoMate Robot - Uses a chip based technology for stable sample infusion at nano litre flow rate.
         
     
TSQ Vantage-Agilent 1290 UHPLC (LC-MS) - The Thermo Scientific TSQ Vantage uses precision hyperbolic quadrupoles, which provide the best transmission and peak shape, the detection technology and ion source results. It is the most sensitive triple quadrupole instrument. Agilent’s 1290 infinity series UHPLC system interfaces to the mass spectrometer for the automated LC-MS/MRM analysis.      
Thermo LTQ Orbitrap Discovery - This MS with its mass resolution of 30,000 at 400 m/z and mass accuracy of 1-2 ppm is optimized for high throughput proteomics experiments. It has two fragmentation techniques, CID and PQD, associated with and is an excellent tool for labeled and label-free quantitation.
