Workshop on 16S Metagenomics by UNCODED Premas kit using Illumina Next-Generation Sequencing
This hands-on workshop provides comprehensive training in 16S rRNA gene-based microbial community profiling using state-of-the-art Illumina next-generation sequencing (NGS) technology and the high-performance Uncoded by Premas 16S Metagenomics Kit.
Participants will gain practical experience in every stage of the 16S metagenomics workflow:

- Sample preparation and library construction using the Uncoded by Premas 16S kit (targeting V3–V4 or full-length regions with optimized primers and bead-based workflow)
- Illumina sequencing on MiSeq (2 × 300 bp paired-end chemistry for maximum taxonomic resolution)
- Quality control, demultiplexing, and preprocessing of raw FASTQ files
- Understanding on complete bioinformatics analysis workflow including:
- Read merging, filtering, and denoising (DADA2)
- OTU/ASV clustering and chimera removal
- Taxonomic assignment using curated databases
- Diversity analysis (alpha and beta diversity)
- Statistical testing and visualization
- Interpretation of microbial community structure, differential abundance, and functional prediction.
The workshop is designed for researchers, students, and professionals in microbiology, ecology, clinical research, and environmental sciences who want to independently perform end-to-end 16S metagenomics studies. No prior NGS or bioinformatics experience is required; basic command-line familiarity is helpful. By the end, participants will be able to confidently design experiments, generate high-quality 16S libraries with the Uncoded kit, run Illumina sequencers, and perform reproducible bioinformatics analysis to answer biological questions about microbial diversity and function.
