NCBS/C-CAMP MS Facility

Proteomics / Protein Characterization

Towards the beginning of this century, mass spectrometry- based proteomics has emerged as a powerful tool for large scale screening, identification and quantitation of proteins from complex biological samples. Moving with this trend, a mass spectrometry (MS) facility was formed at NCBS with an aim to provide researchers state-of-the-art techniques to characterize biomolecules with an emphasis on proteins and peptides.

Services
Currently, the following services are offered from this facility:
 1. Molecular mass measurement of intact proteins and peptides
 2. Identification of 1 D and 2 D gel separated proteins
 3. In-solution digestion and characterization of proteins
 4. De novo sequencing of peptides

Quantitative Metabolomics

Low-molecular weight, endogenous metabolites play critical roles in diverse physiological and pathological processes by participating in biological reactions and that are characterized by various necessary biological functions. By quantifying the concentration changes of small molecules from the biological system in a given condition, we can learn a lot about these biological roles of small molecules or metabolites.


Advances in liquid chromatography-mass spectrometry (LC-MS) techniques amongst others have enabled the identification and quantification of metabolites which are present in pictogram to nanogram range in the biological system. Through our facility at C-CAMP, we are developing mass spectrometry based methods (LC-MS/SRM) for quantifying small molecules (categorizing based on chemical classes) from various biological fluids. As a first attempt, we have successfully developed a method to quantify sixteen neurotransmitters from Planaria extract. This quantification method is now being extended to various other biological matrices. We are also developing methods for the other classes of molecules such as amino acids and derivatives, carnitines, estrogen metabolites etc.,

1. Full scan analysis of known or purified compounds (Solid/Liquid)
2.Product ion scan (MS/MS) analysis of known or purified compounds (Solid/Liquid)
3.Analysis of compounds in biological matrix
4.Method development for specific metabolites to determine the absolute quantification
    - Quantification of Neurotransmitters from biological matrices
5.Analysis and purification using UHPLC system
